Althoughthemixedisomersof5(6)-TAMRAisapreferred,routinelyusedredfluorescentdyeforstainingproteins,labelingpeptidesandnucleotideswithTAMRAmixedisomersmightbetroublesomeduetosignificantsignalbroadeninginHPLCpurification.PeptidesandnucleotideslabeledwithasingleisomerTAMRAusuallygivebetterresolutioninHPLCpurificationthatisoftenrequiredintheconjugationprocesses.
Insummary,theintroductionofapicolylmoietyintoanazideprobeleadstoasubstantialincreaseinthesensitivityofalkynedetection.Thiswillbeofspecialvalueforthedetectionoflowabundancetargetsorwheresignificantincreaseinsignalintensityisdesired.
